Background
VE-Cadherin (vascular endothelial cadherin, CDH5) is a 140-kDa transmembrane glycoprotein that forms the core of endothelial adherens junctions, where its calcium-dependent homophilic interactions between extracellular EC1-EC5 repeats create a semi-permeable barrier between blood and tissues; its cytoplasmic tail recruits β-catenin, plakoglobin and p120-catenin, linking the junction to the actin cytoskeleton and enabling transmission of shear-stress signals that regulate angiogenesis, vascular remodeling and leukocyte extravasation, while phosphorylation of Y685 and Y658 by Src, VEGFR2 or Tie-2 and subsequent internalization of the complex transiently increases permeability during inflammation, tumor metastasis and atherosclerosis, making VE-cadherin a key therapeutic target in vascular pathologies.
