  • Bewegen – Verplaats een van je bouwers naar een aangrenzend vakje. Je mag je bouwer op hetzelfde niveau verplaatsen, één niveau omhoog stappen of een willekeurig aantal niveaus omlaag.
  • Bouwen – Bouw vervolgens een bouwlaag op een vakje naast de bouwer die je net hebt verplaatst. Als je bouwt op de derde verdieping, plaats je in plaats daarvan een koepel, waardoor dat vakje uit het spel wordt gehaald.

Hoe win je? Als één van jouw bouwers de derde verdieping bereikt, win je het spel.

Santorini bevat unieke spelerskrachten die een extra laag toevoegen aan het abstracte basisspel. Er zijn verscheidene thematische goden- en heldenkrachten die elk op hun eigen manier de spelregels veranderen.
