Background
Dicer is a multidomain RNase III enzyme that recognizes double-stranded RNA (dsRNA) or short-hairpin RNA (shRNA), measures ~22 nt from the 3′ overhang, and cleaves both strands to generate small interfering RNAs or microRNAs bearing 2-nt 3′ overhangs; these products are then loaded into Argonaute to form the RNA-induced silencing complex that drives gene silencing, making Dicer the central processor of the RNA interference pathway essential for development, viral defense, and genome stability.
